Deciphering the origin of nuclear speckle-derived DNA using a nuclear matrix protein immuno-fluorescence technique.
A ‘nuclear speckle’ is an inhomogeneous euchromatic foci found in the nucleus of many cells. The DNA in nuclear speckles has been reported to be as a good marker of active transcription by gene expression. However, the origin of nuclear speckle-derived DNA is not clearly understood. In this study, we used a nuclear matrix protein immuno-fluorescence technique to understand the origin of the nuclear speckle-derived DNA. Using antibodies against the nuclear matrix proteins lamin B1 and lamin B2, we found that lamin B1 is present in nuclear speckles and lamin B2 does not. Furthermore, we demonstrated that the nuclear speckle-derived DNA is resistant to nuclease digestion and there is a high level of transcription-activity in the nuclear speckles. Moreover, our results indicate that the nuclear speckles contain a relatively high level of histones H3 and H4, but no H1, in the nuclear matrix. This is a new finding that can be used to further understand the origin of nuclear speckle-derived DNA.
