In vivo effects of phthalate exposure on glucose tolerance, insulin secretion, and lipid metabolism in target organs were investigated using the Zucker obese fatty rat, a genetic model of the metabolic syndrome. Young Zucker rats were exposed to DnBP via diet; older rats were exposed to DEHP via diet or drinking water. Phthalates altered insulin secretion in vivo in Zucker rats, as previously demonstrated by our laboratory. DnBP decreased insulin secretion in younger rats but increased it in older rats. In the liver, phthalate exposure up-regulated HMG-CoA reductase activity, a target enzyme of HMG-CoA and mevalonate. DEHP exposure suppressed lipogenesis and restored fatty acids oxidation in the liver. In vitro, DnBP exposure increased malonyl-CoA decarboxylase activity, a rate-limiting enzyme for fatty acid synthesis. In contrast, DEHP exposure decreased lipogenesis and restored fatty acids oxidation in primary hepatocyte cultures. These results suggest that reduced endogenous sterol esterification by DnBP may have resulted in increased HMG-CoA reductase activity and enhanced insulin secretion.
